【】? Objective? To investigate the expression of LINC01268 and miR-582-5p in pancreatic cancer and their diagnostic value in early pancreatic cancer. Methods? 107 patients with pancreatic cancer (pancreatic cancer group), 80 patients with pancreatitis (benign group) and 50 patients with normal physical examination (control group) admitted to the hospital from January 2022 to March 2024 were selected as research objects. 107 patients with pancreatic cancer were divided into 31 patients with early pancreatic cancer and 76 patients with intermediate and advanced pancreatic cancer. Fluorescence quantitative PCR technology was used to detect the levels of LINC01268 and miR-582-5p, and chemiluminescence method was used to measure the level of carbohydrate antigen 199 (CA199). Multivariate logistic regression analysis was used to analyze the risk factors affecting the occurrence of middle and advanced pancreatic cancer, and ROC curve was drawn to analyze the diagnostic value of LINC01268, miR-582-5p and CA199 levels in early pancreatic cancer. Pearson correlation was used to analyze the correlation between plasma LINC01268, miR-582-5p levels and CA199 in pancreatic cancer patients. Results? The expression level of LINC01268 (2.68±1.04 vs 1.02±0.35) in pancreatic cancer tissues was significantly higher than that in paracancerous tissues, while the expression level of miR-582-5p (0.51±0.13 vs 1.26±0.43) was significantly lower than that in paracancerous tissues(P<0.001). The levels of plasma LINC01268 and CA199 in pancreatic cancer group were significantly higher than those in benign group and control group, while the plasma level of miR-582-5p in pancreatic cancer group was significantly lower than those in benign group and control group (P<0.001). The levels of plasma LINC01268 and CA199 were significantly increased in the middle and advanced pancreatic cancer group, while the levels of plasma miR-582-5p were significantly reduced (P<0.001). The levels of plasma LINC01268 and CA199 were significantly increased in the lymph node metastasis group, while the levels of plasma miR-582-5p were significantly reduced (P<0.001). Multivariate logistic regression analysis showed that increased levels of LINC01268 (OR=4.105, 95% CI: 2.983-12.864) and CA199 (OR=2.790, 95% CI: 2.307-10.116), and decreased levels of miR-582-5p (OR=2.476, 95% CI: 1.915-9.521) were risk factors for advanced pancreatic cancer (all P<0.05). ROC curve analysis showed that the combination of LINC01268≥2.16, miR-582-5p≤0.57 and CA199≥64.82 U/ml had the maximum area under the curve (AUC) 0.925 (95% CI: 0.862-0.990) in the diagnosis of early pancreatic cancer, and its accuracy was 87.6%. Correlation analysis showed that the plasma LINC01268 level in pancreatic cancer patients was positively correlated with CA199 (r=0.825, P<0.001), and the plasma miR-582-5p level was negatively correlated with CA199 (r= - 0.716, P<0.001). Conclusion? In patients with pancreatic cancer, the expression of LINC01268 is increased, and the expression of miR-582-5p is decreased, which is related to tumor stage and lymph node metastasis. The combined detection of LINC01268, miR-582-5p and CA199 has a good diagnostic value for early pancreatic cancer. |
