目的:观察丙酮酸激酶 M2(pyruvate kinase M2,PKM2)和 PD-L1在前列腺癌和良性前列腺增生组织中的的表达,探讨 PKM2 与 PD-L1的相关性。 方法:通过免疫组织化学(Immunohistochemistry,IHC)技术,对58例前列腺癌患者肿瘤组织及58例良性前列腺增生组织切片进行PKM2和PD-L1的表达检测。根据染色强度和阳性细胞百分比,量化两种蛋白的表达水平。同时,收集患者的临床病理资料,包括TNM分期、年龄、淋巴结转移和Gleason评分,分析PKM2和PD-L1的表达与这些参数之间的关系。采用SPSS 26.0统计软件对所得数据进行统计学分析。 结果:PKM2和PD-L1在PCa组织中的表达水平显著高于BPH组织(P<0.05)。进一步分析发现,PKM2和PD-L1的表达水平与PCa的TNM分期和Gleason评分显著相关(P值分别为<0.05;<0.001),即随着肿瘤分期的升高,PKM2和CD276的表达水平亦随之升高。然而,两者的表达与患者年龄、淋巴结转移无关(P>0.05)。此外,统计学分析还揭示了PCa组织中PD-L1的表达与PKM2的表达呈显著正相关,意味着在肿瘤组织中,PKM2表达越高,PD-L1的表达也越倾向于升高。 结论:PKM2 和 PD-L1在前列腺癌中的高表达与肿瘤进展相关。 PKM2 可能通过增加PD-L1的表达而参与免疫调控。 |

Objective: To observe the expression of pyruvate kinase M2 (PKM2) and PD-L1 in prostate cancer and benign prostatic hyperplasia tissues, and to explore the correlation between PKM2 and PD-L1. Methods: Immunohistochemistry (IHC) was used to detect the expression of PKM2 and PD-L1 in tumor tissues of 58 prostate cancer patients and 58 benign prostatic hyperplasia tissues. The expression levels of both proteins were quantified based on staining intensity and percentage of positive cells. Clinical pathological data of patients, including TNM staging, age, lymph node metastasis, and Gleason score, were collected to analyze the relationship between the expression of PKM2 and PD-L1 and these parameters. Statistical analysis was performed using SPSS 26.0. Results: The expression levels of PKM2 and PD-L1 in PCa tissues were significantly higher than those in BPH tissues (P<0.05). Further analysis revealed that the expression levels of PKM2 and PD-L1 were significantly correlated with TNM staging and Gleason score of PCa (P<0.05; <0.001 respectively), indicating that as the tumor stage increased, the expression levels of PKM2 and PD-L1 also increased. However, the expression of both proteins was not associated with patient age or lymph node metastasis (P>0.05). Moreover, statistical analysis also revealed a significant positive correlation between the expression of PD-L1 and PKM2 in PCa tissues, suggesting that higher expression of PKM2 in tumor tissues tended to be associated with increased expression of PD-L1. Conclusion: The high expression of PKM2 and PD-L1 in prostate cancer is associated with tumor progression. PKM2 may be involved in immune regulation by increasing the expression of PD-L1. |
