      摘 要:[目的] 检测外源性IL-17对胃癌细胞生物学特性的影响,探讨其可能的作用机制。[方法] RT-PCR法检测3种胃癌细胞株中IL-17R的表达;MTT法、细胞划痕和Transwell侵袭实验分别检测IL-17对胃癌细胞增殖、迁移和侵袭能力的影响;ELISA实验测定IL-17处理后培养上清中IL-6、IL-8和TNF-α的含量;Western blot技术检测IL-17对通路蛋白Akt、NF-κB及p-Akt、p-NF-κB表达的影响。[结果] IL-17R在3种细胞株均有表达,且在SGC7901细胞中表达最高;外源性IL-17处理胃癌细胞后,其增殖活性无明显变化,但细胞迁移距离明显增大,穿膜细胞数明显增多,培养上清中IL-6和IL-8的分泌水平显著增高,而TNF-α的分泌水平无明显变化;细胞内Akt和NF-κB及p-Akt、p-NF-κB表达水平均显著增高。[结论] 外源性IL-17对胃癌细胞增殖无明显影响,但可明显促进其迁移和侵袭。IL-17可能通过调节PI3K-Akt信号通路上调IL-6和IL-8的分泌,促进胃癌细胞迁移和侵袭

      Abstract:[Purpose] To investigate the effect of IL-17 on biological activity of gastric cancer cells and its related mechanisms. [Methods] RT-PCR was used to analyze the expression of IL-17R in three gastric cancer cell lines. MTT assay was used to detect cell proliferation. Wound healing assay and Transwell assay were used to analyze migration and invasion of SGC7901 cells . ELISA was used to detect the levels of IL-6,IL-8 and TNF-α. Western blot was used to analyze the Akt,NF-κB,p-Akt and p-NF-κB protein expressions. [Results] IL-17R was expressed in all three gastric cancer cell lines and SGC7901 has the highest expression. After treatment by IL-17,the proliferation of gastric cancer cells had no significant changes,however,cells migration distance was markedly increased,the number of transmembrane cells was significantly increased. The levels of IL-6 and IL-8 in the culture supernatant were significantly increased,while there was no difference in the level of TNF-α. The protein expressions of Akt,NF-κB,p-Akt and p-NF-κB were significantly increased. [Conclusion] Exogenous IL-17 has no significant effect on proliferation of gastric cancer cells,but it promotes migration and invasion of gastric cancer cells. IL-17 may upregulate the secretion of IL-6 and IL-8 by regulating PI3K-Akt signaling pathway,result in promoting migration and invasion of gastric cancer cells.
