摘 要:[目的] 检测食管鳞状细胞癌(esophageal squamous cell carcinoma, ESCC)组织中微小RNA 4659b(microRNA-4659b,miR-4659b)和长链非编码RNA XLOC_008370(long non-coding RNA XLOC_008370,lncRNA XLOC_008370)的表达,探讨miR-4659b和XLOC_008370在ESCC发生及发展中的作用机制。[方法] 应用实时荧光定量PCR(real-time quantitative polymerase chain reaction,qRT-PCR)的方法检测miR-4659b和XLOC_008370在50例ESCC组织中的表达情况及相关性。[结果] ESCC组织中miR-4659b的相对表达量显著高于相应癌旁组织[(0.034±0.013) vs (0.005±0.002),t=15.205, P<0.05]。miR-4659b的相对表达量与ESCC患者淋巴结转移有关 [(0.039±0.011)vs(0.023±0.009),t=5.708,P<0.05]。XLOC_008370在ESCC中的相对表达量显著低于相应癌旁组织[(0.0019±0.0012)vs(0.020±0.0146),t=-8.921,P<0.05]。XLOC_008370的相对表达量与ESCC患者淋巴结转移有关[(0.0016±0.0010)vs(0.023±0.0014),t=-2.040, P<0.05],并且XLOC_008370与miR-4659b的相对表达量呈负相关(r=-0.829,P<0.05)。[结论] miR-4659b在ESCC中表达上调而XLOC_008370表达降低,且两者之间有相关性,提示miR-4659b与XLOC_008370可能存在一定的调控作用,这一机制可能在ESCC发生发展中起重要作用。 |

Abstract:[Purpose] To investigate the expressions of microRNA-4659b and long non-coding RNA (lncRNA) XLOC_008370 in esophageal squamous cell carcinoma (ESCC) and their relations to clinicopathological features. [Methods] Real-time quantitative polymerase chain reaction (qRT-PCR) was used to detect the expressions of microRNA-4659b and lncRNA XLOC_008370 in 50 ESCC patients,and their clinicopathological significance was studied. [Results] The expression of miR-4659b in ESCC tumor tissues was significantly higher than that in cancer adjacent tissues [(0.034±0.013) vs (0.005±0.002),t=15.205,P<0.05] and the expression was higher in ESCC with lymph node metastasis than that in non-metastatic ESCC[(0.039±0.011) vs (0.023±0.009),t=5.708,P<0.05]. The expression of XLOC_008370 in ESCC tumor tissues was significantly lower than that in cancer adjacent tissues [(0.0016±0.0010) vs (0.023±0.0014),t=-2.04,P<0.05] and the expression was lower in ESCC with lymph node metastasis than that in non-metastatic ESCC [(0.039±0.011) vs (0.023±0.009),t=5.708,P<0.05]. The relative expression of XLOC_008370 was negatively correlated with miR-4659b(r=-0.829,P<0.05). [Conclusion] The expression of miR-4659b is up-regulated while XLOC_008370 is down-regulated in ESCC,and they are negatively correlated with each other. Results suggest that MiR-4659b may target to regulate XLOC_008370,and it might be involved in the development of ESCC. |
