      摘 要:[目的] 探讨乳腺癌组织CC类趋化因子5(CC chemokine ligand 5,CCL5)与上皮间质转化(epithelial-mesenchymal transition,EMT)之间的相关性,探索抑制乳腺癌浸润转移的新途径。[方法] 应用免疫组织化学技术检测101例浸润性乳腺癌组织中CCL5、基质金属蛋白酶-9(matrix metalloproteinase 9,MMP-9)、钙黏附蛋白E(E-Cadherin)、钙黏附蛋白N(N-Cadherin)、波形蛋白(Vimentin)的表达,研究乳腺癌浸润转移关键分子CCL5、MMP-9表达与EMT之间的关联性,分析CCL5促EMT发生的可能途径。[结果] CCL5与MMP-9的表达率和EMT的发生率分别为34.7%(35/101)、55.4%(56/101)和41.6%(42/101)。乳腺癌CCL5表达与淋巴结转移相关(P<0.05),与其他临床病理特征无关(P>0.05)。Spearman分析显示乳腺癌组织中CCL5与MMP-9表达呈正相关(r=0.276,P=0.005)。二元Logistic回归分析显示,只有MMP-9与EMT有相关性(OR=4.697,P=0.001)。[结论] 乳腺癌组织中CCL5的表达可促进EMT的发生,其作用途径可能与上调MMP-9有关。

      Abstract:[Purpose] To investigate the correlation between the expression of CCL5 and epithelial-mesenchymal transition(EMT) in breast cancer and to explore the new approach of invasion and metastasis in breast cancer. [Methods] The expressions of CCL5,MMP-9,E-Cadherin,N-Cadherin and Vimentin protein were detected in the cancer cells of 101 cases with invasive breast cancer by immunohistochemistry. The relationships between the expressions of CCL5,MMP-9,and epithelial-mesenchymal transition(EMT) were analyzed,possible way of CCL5 promotes EMT was analysised. [Results] The expression rates of CCL5,MMP-9,and the rate of EMT were 34.7%(35/101),55.4%(56/101),and 41.6%(42/101),respectively. The expression of CCL5 protein in breast cancer was correlated with lymph node metastasis(P<0.05),but not with other clinicopathological features(P>0.05). Spearman analysis showed that the expression of CCL5 was correlated with the expression of MMP-9(r=0.276,P=0.005). Logistic regression analysis showed that only MMP-9 induced EMT (OR=4.697,P=0.001). [Conclusion] The expression of CCL5 is correlated with EMT in breast cancer,maybe by up-regulation the expression of MMP-9.
