      摘 要:[目的] 研究电压门控质子通道蛋白Hv1在结肠癌组织中的表达及其临床病理学意义。[方法] 回顾性分析2004年5月至2008年5月的87例结肠癌患者临床病理学资料;免疫组化检测87例结肠癌组织和30例癌旁正常组织石蜡标本中Hv1表达。Cox风险回归模型回归分析影响结肠癌患者预后的独立影响因素。[结果] Hv1在结肠癌组织中呈阳性表达,在癌旁正常组织中呈阴性表达。Hv1表达水平与患者年龄、肿瘤大小、肿瘤T分期、淋巴转移、远处转移及临床分期等临床病理因素之间有显著性相关(P<0.05)。Kaplan-Meier生存分析显示,高表达Hv1的结肠癌患者预后差,与Hv1低表达的患者相比,Hv1高表达的患者中位生存期明显缩短(46个月 vs. 53个月,P<0.05),无复发中位生存时间也明显缩短(36个月 vs. 49个月,P<0.05)。COX回归分析显示Hv1表达是结肠癌患者总生存率(P=0.014)和无复发生存率(P=0.037)的独立影响因素。[结论] 电压门控质子通道蛋白Hv1在结肠癌组织中阳性表达,是结肠癌患者预后生存的独立影响因素,是治疗结肠癌药物研发的潜在靶点。

      Abstract:[Purpose] To investigate the expression and clinicopathological significance of voltage-gated proton channel Hv1 in colon cancer tissues. [Methods] Clinicopathological data of 87 patients with colon cancer hospitalized from May, 2004 to May, 2008 were retrospectively reviewed. Expression of voltage-gated proton channel Hv1 in 87 cases with colon cancer tissues and 30 cases with adjacent normal tissues was determined by immnunohistochemistry (IHC). Cox proportional hazard regression model analysis was performed to analyze the independent prognostic factors of patients with colon cancer. The influence of Hv1 expression on the overall survival and recurrence-free survival of patients with colon cancer was analyzed by Kaplan-Meier method with Log-rank test. [Results] Hv1 protein was positively expressed in colon cancer tissues , while negatively expressed in adjacent normal tissues. Hv1 expression in colon cancer tissues significantly correlated with patients’ age, tumor size, T stage, lymph node status, distant metastasis and clinical stage. Cox proportional hazard regression analysis indicated that Hv1 expression was independent prognostic factor of overall survival (P=0.014) and recurrence-free survival(P=0.037). Kaplan-Meier survival analysis showed that patients with high Hv1 expreesion had poorer prognosis than those with low Hv1 expression. Compared with patients with low Hv1 expression, patients with high Hv1expression had a shorter median survival (46 months vs 53 months, P<0.05) and shorter recurrence-free survival (36months vs 49months,P<0.05). [Conclusion] Voltage-gated proton channel protein Hv1, positively expressed in colon cancer tissues, is independent prognostic factor of patients with colon cancer. Hv1 is a promising potential target for developing new methods of tumor therapy.
