| 摘 要：肿瘤细胞原代培养技术可为日益深入的肿瘤研究及治疗提供充足有效的细胞来源，是癌症在体外研究的重要手段，与体内研究相辅相成。但目前肿瘤细胞原代培养成功率不高，许多问题还需继续探讨与研究。全文综述目前国内外肿瘤原代培养方法、条件及冻存注意事项，包括：肿瘤组织类型与处理、细胞培养及纯化方法、原代肿瘤细胞生物特性检测、肿瘤细胞冻存条件等，综合探讨各方法的优缺点。比较获得更有效的肿瘤细胞原代培养方法，将其运用于探索肿瘤发生机制、肿瘤细胞治疗及药敏试验，推进肿瘤治疗的进程。 |

| Abstract：Primary culture techniques can provide adequate and effective source of tumor cells for the cancer deepening research and treatment，which is an important methodology for vitro cancer research. Furthermore，it is the supplementary to vivo studies. But the success rate of tumor cells in primary culture is not high，and many questions need to be figure out with sustained exploration. This paper reviews the current tumor primary culture methods，conditions and frozen precautions，including 1) tumor tissue type and treatment，2) methodology of primary culture and purification，3) detecting the biological characteristics of primary tumor cells，4) freezing conditions. The paper will give a comprehensive discussion of various advantages and disadvantages of the method above-mentioned. It is conducive to obtain more effective primary culture methods to explore the tumor mechanism，tumor therapy and drug susceptibility testing，which will promote the process of tumor therapy，and bring the gospel to cancer patients. |
