摘 要:[目的] 通过检测胃癌患者外周血单核细胞中CD133的表达,探讨其与临床病理特征的关系。[方法] 于术前抽取30例胃癌患者、15例胃溃疡患者、15名健康志愿者外周静脉血,密度梯度离心法分离单核细胞。半定量反转录聚合酶链反应检测CD133 mRNA表达水平。分析胃癌患者外周血CD133 mRNA表达与临床病理特征的关系。[结果] 胃癌患者外周血中CD133 mRNA表达水平明显高于胃溃疡患者和健康志愿者(0.2804±0.1835 vs 0.0984±0.1321,t=6.724,P<0.001;0.2804±0.1835 vs 0.0282±0.0597,t=-7.327,P<0.001)。CD133 mRNA表达与肿瘤最大直径、淋巴结转移、TNM分期有关(P<0.05),而与性别、分化程度、淋巴管浸润、血管浸润、浸润深度无关。Spearman相关分析显示,CD133 mRNA表达与淋巴结转移呈正相关(P<0.01)。[结论] 胃癌患者外周血中CD133 mRNA的表达与胃癌的发展、转移及预后密切相关。 |

Abstract:[Purpose] To detect the expression of CD133 mRNA in peripheral blood mononuclear cells of patients with gastric cancer and to investigate its relationship with clinicopathological features.[Methods] Peripheral venous blood of 30 cases with gastric cancer,15 cases with gastric ulcer and 15 cases of healthy volunteers before surgery was taken. Mononuclear cells were isolated by density gradient centrifugation. The expression of CD133 mRNA was detected by semi-quantitative RT-PCR,and its relationship with clinicopathological features was analyzed.[Results] The expression levels of CD133 mRNA in peripheral blood of patients with gastric cancer(0.2804±0.1835) were significantly higher than that in patients with gastric ulcer(0.0984±0.1321) and healthy volunteers (0.0282±0.0597)(all P<0.001).The expression of CD133 mRNA in the peripheral blood of patients with gastric cancer before operation was related to tumor diameter,lymph node metastasis and TNM staging (all P<0.05),but not related to gender,differentiation,lymphatic vessel invasion,vascular invasion and infiltration depth. Spearman correlation analysis showed that there was a positive correlation between the expression of CD133 mRNA in the peripheral blood and lymph node metastasis of patients with gastric cancer(P<0.01).[Conclusions] The expression of CD133 mRNA in peripheral blood is closely related with development,metastasis and prognosis of patients with gastric cancer. |
