      摘 要:[目的] 初步研究半抗原2,4-二硝基氟苯(2,4-DNFB)在体内外对肝细胞癌的抑制作用。[方法] 用不同浓度的DNFB、氟尿嘧啶(5-Fu)(对照组)分别处理SMMC-7721肝癌细胞株72h,用MTT法测定72h细胞存活率,并计算50%和90%抑制浓度(IC50和IC90)。经H22肝癌细胞株皮下种植成功的18只昆明鼠随机分成3组:DNFB组、无水乙醇(Ethanol)组、磷酸盐缓冲液(PBS)组(对照组),并分别瘤内注射100μl的DNFB、Ethanol和PBS,每周2次,共4次。瘤内注射结束2周后剥离肿瘤称重,计算抑瘤率。[结果] DNFB和5-Fu对SMMC-7721肝癌细胞株的IC50分别为32.22μmol/L和3.62μmol/L;IC90分别为49.53μmol/L和>200μmol/L。DNFB组、无水乙醇组和PBS对照组最终的瘤重分别为0.97±0.53g、1.55±0.76g和3.79±0.75g。与PBS对照组比较,DNFB组和无水乙醇组的抑瘤率分别为74.43%(P=0.00004)和59.20%(P=0.00045),DNFB组和无水乙醇组之间的差异无统计学意义(P=0.16)。[结论] DNFB可在体内外抑制肝癌细胞的生长。

      Abstract:[Purpose] To investigate inhibitory effect of hapten 2,4-dinitrofluorobenzene (2,4-DNFB) on hepatocellular carcinoma (HCC) in vitro and in vivo. [Methods] SMMC-7721 cells were treated with DNFB and 5-fluorouracil (5-Fu) (control group) under different concentrations for 72 hours. The methyl thiazolyl tetrazolium (MTT) method was used to determine survival rate of HCC cells,and to calculate IC50 and IC90. Eighteen Kunming mice,of which H22 cells were successfully implanted into subcutaneousness,were randomly divided into three groups,DNFB group,Ethanol group and PBS group(control group),and 100μl DNFB,ethanol and PBS were intra-tumor rally injected in each group,twice a week for two weeks,respectively. After two weeks,the tumor were weighted and the rates of tumor inhibition (RTI) were calculated. [Results] Contrations of DNFB and 5-Fu to reach IC50 of SMMC-7721 cells were 32.22μmol/L and 3.62μmol/L respectively,and those to reach IC90 were 49.53μmol/L and >200μmol/L,respectively. The tumor weights in DNFB group,Ethanol group and PBS control group were 0.97±0.53g,1.55±0.76g and 3.79±0.75g,respectively. Compared with PBS control group,the RTI was 74.43% in DNFB group (P=0.00004) and 59.20% in Ethanol group(P=0.00045), but there was no statistically difference between DNFB group and Ethanol group(P=0.16). [Conclusion] Hapten DNFB can inhibit HCC growth in vitro and in vivo.
